If, for example, I shoot a doe and tag it with my muzzleloader license, can I purchase a second muzzleloader license in order to continue hunting for a buck? Thanks in advance. Steve

No. Under a muzzleloader license, you may take either a buck or a doe. You are allowed only one muzzleloader license per year. I would suggest that you purchase a bonus antlerless license which you can use any method in season to take a doe and then you would still have your muzzleloader license for the buck.
